Directed Evolution of Stereoselective Biocatalysts
INTRODUCTION Stereoselective chemical reactions have become extremely important in both academic and industrial settings for the synthesis of useful molecules. Catalytic reactions are of particular importance due to their capacity to effect otherwise impossible transformations using substoichiometric quantities of the catalyst. Traditionally, transition metal catalysts have played a central rol...
متن کاملAffinity-labelling of the thyrotropin receptor
Thyrotropin (TSH) has been coupled to the photoactive heterobifunctional reagent N-hydroxysuccinimidyl 4-azidobenzoate (HSAB) and the properties of the product (HSAB-TSH) investigated. Preparations of HSAB-TSH containing two molecules of HSAB per molecule of TSH were used in most experiments and these preparations retained about 40% of the original receptor-binding activity of the TSH. HSAB-TSH...
متن کاملHydroxyl-Directed Stereoselective Diboration of Alkenes
An alkoxide-catalyzed directed diboration of alkenyl alcohols is described. This reaction occurs in a stereoselective fashion and is demonstrated with cyclic and acyclic homoallylic and bishomoallylic alcohol substrates. After oxidation, the reaction generates 1,2-diols such that the process represents a method for the stereoselective directed dihydroxylation of alkenes.
متن کاملMetal free stereoselective synthesis of functionalized enamides.
An efficient and expeditious DABCO-mediated synthesis of functionalized enamides from alkenes is delineated. The reaction proceeds through an unprecedented cascade involving an Aza-Michael addition/α-bromination/elimination and a Morita-Baylis-Hillman type reaction to generate functionalized enamides in a regio- & stereoselective fashion.
متن کاملAffinity labelling of 5'-nucleotidases with 5'-p-fluorosulphonylbenzoyladenosine.
5'-Nucleotidases play an important role in the metabolism of nucleosides; for example, the hydrolysis of AMP generates adenosine, which can modulate a variety of cellular functions. We have used the membrane-bound AMPase from chicken gizzard and a secreted form of these enzymes to analyse their modification by the substrate analogue 5'-p-fluorosulphonylbenzoyladenosine (5'-FSBA). 5'-FSBA irreve...
